Virtual Issue: Most Original and Most Significant Scientific Findings

This quarter we feature Bruno Hancock’s comments for the paper he chose: “Physical Stability and Dissolution of Lumefantrine Amorphous Solid Dispersions Produced by Spray Anti-Solvent Precipitation”

In this paper a new method of manufacturing amorphous solid dispersions (ASDs) is studied. The proposed anti-solvent precipitation process has the potential benefit of needing less solvent than a conventional spray-drying approach. The authors studied several polymeric carriers and compared the manufacturability and dissolution performance of the resulting ASDs. Both the choice of polymer, and the drug-polymer ratio played a crucial role in the solubility enhancement of the model drug substance (lumefantrine). The results of this study are important because they confirm the feasibility of producing ASDs using a simplified manufacturing approach.
